Ipamorelin + GHRP-2

1 Identity

A fixed-ratio two-component article: two short synthetic peptides of the GHRP class, one a pentapeptide and one a hexapeptide, co-lyophilized. The only blend in this batch with no large component — both are sub-kilodalton, both carry beta-(2-naphthyl)alanine, both are C-terminally amidated.

Sequence
Ipamorelin is Aib-His-D-2-Nal-D-Phe-Lys-NH2, five residues; GHRP-2 is H-D-Ala-D-2-Nal-Ala-Trp-D-Phe-Lys-NH2, six residues. Both share the C-terminal D-Phe-Lys-NH2 dipeptide, so the y2 ion is common to both and MS/MS quantitation must be built on precursor-specific or N-terminal fragment ions rather than on the shared C-terminal series.
Molecular formula
Ipamorelin free base C38H49N9O5; mono-acetate C40H53N9O7; bis-acetate C42H57N9O9; mono-TFA C40H50F3N9O7. GHRP-2 free base C45H55N9O6; mono-TFA C47H56F3N9O8; dihydrochloride C45H57Cl2N9O6. Both carry three basic sites and both are routinely shipped as multi-protonated salts, so counterion is determined per component and the gross-weight figure is never used as a content proxy. Average mass (IUPAC 2021) Ipamorelin 711.868 Da; GHRP-2 817.992 Da.
Monoisotopic mass
Ipamorelin 711.3857 neutral, [M+H]+ 712.3930, [M+2H]2+ 356.7001; GHRP-2 817.4275 neutral, [M+H]+ 818.4348, [M+2H]2+ 409.7210. The 106.124 Da separation is adequate for resolution, but the doubly charged ions sit only 53.02 m/z apart and both fall in the same window as common matrix ions, so the identity assignment is made on the singly charged species or on accurate mass, not on the 2+ envelope alone.
Salt / variant note
At a nominal 5 mg + 5 mg fill, 10 mg total, the 1.0 : 1.0 mass ratio is a molar ratio of 1.00 : 0.87 — 7.02377 against 6.11253 micromol. This is the one blend in this batch where the mass ratio and the molar ratio are close, because the two components differ by only 106.124 Da. The naphthylalanine family runs ipamorelin 711.868, GHRP-2 817.992, GHRP-6 873.032 and hexarelin 887.06, all of which are routinely substituted into blends. Ipamorelin free-acid and des-amido forms sit at a 0.98 Da offset, unresolvable on a nominal-mass instrument. Counterion variants — acetate, bis-acetate, trifluoroacetate, dihydrochloride — are determined per component and measured. There is no standard ratio; 5 mg + 5 mg is stated nominal only.

Published literature exists for each component and each body of work is graded on its own. The ipamorelin literature is preclinical: a small number of company-sponsored phase 1 and phase 2 studies were registered and completed in the 2000s, the program was discontinued, and no product has been approved and no phase 3 dataset exists. The GHRP-2 literature includes human studies published under PMID 17609397 and PMID 23079545, and GHRP-2 is the active moiety of a medicine registered in Japan.

4 Storage & specification

Storage
A white to off-white co-lyophilized solid in a screw-cap amber borosilicate vial with a PTFE-lined closure — a laboratory-chemical presentation, non-sterile, with no sterility claim, and not a stoppered and crimped injection vial. Store at -20 degrees C plus or minus 5 degrees C, desiccated, tightly closed and protected from light in amber glass or a foil overwrap: both components carry a naphthalene ring and GHRP-2 additionally carries a tryptophan indole, so every chromophore in this article is photolabile. Both salts are hygroscopic, and sealed vials are equilibrated to ambient temperature before opening.
Shelf life
Provisional 24 months at -20 degrees C plus or minus 5 degrees C, desiccated and protected from light, from QA release — both component intervals being provisional 24 months, so the shorter-of rule returns the same figure here, the one blend in this batch where it does. The blend runs its own protocol and inherits no component study, with a mandatory 6-month interim pull on the first three lots. Trended attributes are per-component net content, the measured ratio, oxidation of the GHRP-2 indole, C-terminal amide hydrolysis on both components, diketopiperazine formation, water, counterion and any new peak above 0.10 percent.
