IGF-1 (human, full length)
Also indexed as Mecasermin (INN), Increlex, recombinant human IGF-1, rhIGF-1, somatomedin C, IGF-I
1 Identity
Single-chain polypeptide, 70 residues, three intramolecular disulfides, non-glycosylated. E. Coli-expressed. Not to be confused with mecasermin rinfabate (iPlex), a separate rhIGF-1/rhIGFBP-3 complex withdrawn from the US market.
- Sequence
- GPETLCGAELVDALQFVCGDRGFYFNKPTGYGSSSRRAPQTGIVDECCFRSCDLRRLEMYCAPLKPAKSA (70 residues; UniProt P05019 residues 49-118, confirmed residue-for-residue against PDB 1GZR chain A). Disulfide connectivity in mature numbering: Cys6-Cys48, Cys18-Cys61, Cys47-Cys52.
- Molecular formula
- C331H512N94O101S7 (oxidized, three disulfides — the correct species for the folded protein).
- Average mass
- 7648.714 Da average.
- Monoisotopic mass
- 7643.58624 Da monoisotopic neutral; [M+H]+ 7644.59352.
- Salt / variant note
- IGF-1 LR3 is an 83-residue analogue carrying an Arg3 substitution and a 13-residue N-terminal extension, about 9,111 Da, and a different molecule. IGF-1 des, also recorded here, is des(1-3)IGF-1, the 67-residue truncation. This article is the 70-residue wild-type and is neither of them. Also IGF-2, insulin, proinsulin and INSL peptides, all sharing the insulin-superfamily disulfide architecture. Sellers routinely file all of these under "IGF-1".

3 Primary sources & evidence
Sequence and connectivity are settled and agree across two independent primary sources, UniProt P05019 and PDB 1GZR. The formula and both masses follow from that sequence.
