Human Relaxin-2

Also indexed as Relaxin-2, H2 relaxin, RLN2

1 Identity

Two-chain peptide hormone, A chain 24 residues and B chain 29 residues, held together by two interchain disulfides with one intrachain disulfide in the A chain. Insulin-superfamily architecture. Not a linear peptide, and not synthesizable as one without a directed folding step. Serelaxin is the recombinant pharmaceutical form.

Sequence
A chain 24 residues, B chain 29 residues. Disulfide connectivity A10-B10, A15-B22 interchain and A11-A16 intrachain. The connectivity is the molecule: a correctly composed preparation with scrambled disulfides has the same mass and is not this article.
Molecular formula
C256H408N74O74S8 for the correctly folded two-chain molecule.
Average mass
Approximately 5963 Da for the folded two-chain molecule (derived).
Monoisotopic mass
Not available from an authoritative source.
Salt / variant note
B7-33, also recorded in this catalog, is a single-chain relaxin analogue, not relaxin, and the two must never be reconciled to one record. Also relaxin-1 and relaxin-3, INSL3 and INSL5, and insulin and IGF-1 and IGF-2, which share the architecture. Note the internal collision: this catalog holds IGF-1 full length, IGF-1 LR3 and Human Relaxin-2, all insulin-superfamily, all disulfide-dependent, and all defeated by the same analytical shortcut.

3 Primary sources & evidence

The index reports the design and provenance of the literature, not a conclusion about effect.

Structure and connectivity are settled in the literature. The evidence from the clinical program is negative and is not reproduced here.

4 Storage & specification

Storage
Lyophilized, -20 degrees C or below, desiccated, protected from light. Reconstituted material at 2 to 8 degrees C for short periods only and never re-frozen; aggregation on freeze-thaw is the dominant failure mode for folded proteins, and it is not visible to the eye.
Shelf life
Not assignable without measured stability data on the folded form.

This record reports identity, specification and study design. It does not state what the article does in a human body. Supplied under the caution: “CAUTION: Contains a new drug for investigational use only in laboratory research animals or for tests in vitro. Not for use in humans.
