CJC-1295 (DAC:GRF)

Also indexed as CJC-1295, DAC:GRF, CJC-1295 with DAC, ConjuChem CJC-1295

1 Identity

GHRH analogues. 'CJC-1295 no DAC' is not this molecule - it is Modified GRF (1-29), which has its own record. [D-Ala2, Gln8, Ala15, Leu27]-GHRH(1-29)-Lys30(3-maleimidopropionyl)-NH2. CAS 446262-90-4.

Sequence
H-Tyr-D-Ala-Asp-Ala-Ile-Phe-Thr-Gln-Ser-Tyr-Arg-Lys-Val-Leu-Ala-Gln-Leu-Ser-Ala-Arg-Lys-Leu-Leu-Gln-Asp-Ile-Leu-Ser-Arg-Lys30(N-epsilon-3-maleimidopropionyl)-NH2.
Molecular formula
C165H269N47O46
Average mass
3647.25 Da (C165H269N47O46).
Monoisotopic mass
3645.0155 Da neutral. Working ions [M+3H]3+ m/z 1216.0124; [M+4H]4+ m/z 912.2612.
Salt / variant note
(1) the central substitution - 'CJC-1295 without DAC', which is Modified GRF (1-29), C152H252N44O42, 3367.954 avg / 3365.89358 mono. It is 279.296 Da lighter and is sold under the same head trade name by most of the same sellers, sometimes on the same product page behind a dropdown. This is a large, unambiguous mass difference fully detectable by arithmetic on any competent instrument, which means a lot substituted this way and shipped with a real certificate proves the certificate was never read. (2) ring-opened maleamic acid degradant - hydrolysis of the maleimide ring gives C165H271N47O47, 3665.265 avg / 3663.02605 mono, +18.011 Da. This is the principal impurity, it forms on storage and on any aqueous handling above neutral pH, it is chromatographically close to the parent so a shallow gradient is required to see it, and it is thiol-UNREACTIVE: a fully ring-opened lot still shows the right amino-acid analysis and near-parent retention while being inert as a conjugating reagent. (3) des-amido (C-terminal free acid) - C165H268N46O47, 3648.234 avg / 3645.9995 mono, +0.984 Da, indistinguishable at unit resolution. (4) thiol adducts formed before release - the maleimide adds any free thiol present in workup; a glutathione adduct is +307.0838 Da monoisotopic and a cysteine adduct +121.0197 Da. (5) the all-L stereoisomer at D-Ala2: identical formula, identical exact mass to every decimal place, mass-silent, and the reason P3 is mandatory. (6) An acyl group placed on the Lys30 alpha-amine rather than the epsilon-amine - a different molecule at the same mass and on the same amino-acid analysis.

3 Primary sources & evidence

The index reports the design and provenance of the literature, not a conclusion about effect.

A small human literature exists and is unusual in this catalog for being sponsor-run and peer-reviewed: phase 1 work in healthy adults from the originating company was published in the Journal of Clinical Endocrinology and Metabolism in 2006 (Teichman and colleagues), alongside preclinical reports. No phase 2 or phase 3 result has been published. Clinical development was discontinued by the sponsor in 2006. No current registered trial appears in ClinicalTrials.gov as of August 2026. This is a description of what has been published and registered, by study type and identifier.

4 Storage & specification

Storage
Lyophilized powder, sealed under nitrogen in an amber 5 mL borosilicate serum vial. Labeled condition -20 degrees C plus or minus 5 degrees C, desiccated, protected from light, with the Additional label statement that the material must not be brought into contact with any free thiol or reducing agent. The aqueous stability window for the maleimide is pH 6.0 to 7.0; above pH 7.5 maleimide ring hydrolysis is rapid and the material degrades to an unreactive form with no visible change. This handling constraint is printed on the vial label, not left to the technical note.
Shelf life
12 months at -20 degrees C plus or minus 5 degrees C, provisional - deliberately half the class default, because maleimide ring hydrolysis proceeds in the solid state at measurable rates and is the retest-limiting attribute rather than peptide backbone degradation. Protocol: 0, 1, 3, 6, 9 and 12 months at the labeled condition with maleamic acid content and thiol-reactivity titration at every point, plus a 3-month 25 degrees C / 60% RH arm to establish the excursion tolerance for shipping.
