How to read a certificate of analysis

A certificate reports several percentages and they don't mean the same thing. Only some tell you how much peptide is in the vial. Here's a made-up certificate in the usual layout — not any one supplier's, so it reads the same whoever your vial came from.

Any teal line gives you the peptide mass. Peptide content in mg is the most direct — enter it as mg. Assay and net peptide content are percentages of the label — enter them as %. Here all three agree: 8.20 mg out of a 10 mg vial.
The amber line does not. HPLC purity is an area percentage of the chromatogram — how much of what's there is the right molecule, not how many mg are there. A vial can be 99.2% pure and still hold only 8.2 mg, because counterion and water are mass that isn't an impurity peak. Never enter a purity % as peptide content.
⚠ No COA? Be careful. A vial with no certificate of analysis has nothing behind its label — no independent check of what the compound is, how pure it is, or how much is actually in the vial. Think hard about whether that vial belongs in your research at all.

If you proceed anyway, use the label amount — the answer is then “nominal,” meaning based on the label alone, which is what every other calculator silently does.
